I ran a gel today (pictured above) that shows that I successfully made a 147kb deletion with the use of TALENs and it was passed from the injected zebrafish to their progeny. Each well (apart from the ladder) displays individual digested embryos being tested for the 147kb deletion, which we are able to test this through PCR. When a band shows up, the deletion has been made because 147kb is not amplifiable.  This was important because:
This is currently (based on the reading I’ve done) the biggest TALEN made deletion to go germline
This is the creation of a second mutant allele for the same long-non coding RNA that has been shown that when mutated, the fish get a phenotype of heritable retinoblastoma.
So basically I’m a woman, a latina and a junior in college and I just did awesome discovery genetic based science.
